Transcriptome and metagenome sequencing of Magallana gigas exposed to low pH and sodium molybdate conditions

Metagenome and transcriptome sequencing to study the effects of ocean acidification on Pacific oyster (M. gigas) development. Also examined the effect of sodium molybdate on oyster growth and microbiome composition. Larval / juvenile oysters were collected at multiple time points. ZymoBIOMICS DNA Miniprep Kits were used to extract total DNA, which was sent to the Integrated Microbiome Resource at Dalhousie University (Halifax, Nova Scotia) for metagenomic PCR amplification and sequencing. Sequencing libraries were prepared by targeting the V4-V5 regions of the 16s rRNA gene for amplification using the the 515F (GTGYCAGCMGCCGCGGTAA) and 926R (CCGYCAATTYMTTTRAGTTT) primer set. Sequencing was performed on the Illumina HiSeq platform. Total RNA was isolated using a TRIZOL Reagent (Life Technologies) protocol. RNA was processed for Tag-Sed using the QuantSeq 3' mRNA-Seq Library Prep Kit FWD for Illumina (Lexogen). Sequencing was performed on the Illumina HiSeq platform.
