Gene expression under high salt stress for coconut

Coconut has a redundant gene pool to flourish under salt stress conditions in particular the retention of genes resulting from the WGD p and the high genome size with a high TE content is a favourable context for gene innovation (e.g. sub or even neo-functionalization events) regarding abiotic stress tolerance. Four known pathways (ABA, ABA independent, ROS, Calcium/PLC and SOS) are present and expressed. Several strategies are in place: (moderate) high constitutive expression with a few number of gene candidates, moderate expression with a higher number of gene candidates, expression regulation of gene candidates. Overall design: A Salt stress experiment was conducted on coconut palms seedlings, weekly irrigated on Murashige and Skoog (MS) culture medium and then enriched with 200 mM NaCl (time 0). After four hours the osmotic stress reached the leaves but not yet the cation stress after six days the osmotic stress was still present in the leaves and the ionic stress could be felt at seven days, a second irrigation of MS with 200 mM was performed after ten days the osmotic and cation stresses should be present in the leaves (after two irrigation there was a NaCl accumulation).Varieties used are Hainan Tall (DB) and Aromatic Coconut (XS). Sampling at 0 time point were used as a control, samples were taken at 4hr, 6day and 10 days after 200 mM NaCl treatment. The experiments were done in three replications but one to two samples were missing for 10 days sampling.
