Identification of novel target genes for Nrf2 in Mytilus coruscus.

Nrf2 plays a pivotal role in regulating oxidative stress, and in mollusks, understanding how Nrf2 governs the expression of new target genes is crucial in the context of oxidative stress. In this study, we have identified solute carrier family 35 member E2 (SLC35E2) as a novel Nrf2 target gene that counteracts the oxidative stress response induced by Bap.Initially, we confirmed the Nrf2-SLC35E2 interaction through dual luciferase and qRT-PCR assays, demonstrating that Nrf2 both targeted and inhibited SLC35E2 expression. Subsequently, we further substantiated Nrf2's involvement in BAP-induced oxidative stress by assessing enzyme activity.Therefore, our study provides valuable insights that can contribute to future research on the environmental adaptation and stress responses of mollusks, signifying its significant importance. Overall design: To investigate novel target genes regulated by Nrf2 and their involvement in oxidative stress responses, we conducted chromatin immunoprecipitation sequencing in Mytilus coruscus. Please note that Nrf2 antibody was custom-prepared by Hangzhou Hua'an Biotechnology Co., Ltd and the purified antibody bears the project number TP30282.
