We compare quantitative polymerase chain reaction with colony forming units derived from three different plating medias. PDA, PDA with chloramphenicol and DRBC with chloramphenicol were tested. Colonies were isolated from each media type and whole genome sequenced to identify the different microbes presented with each selection. Each plating medium displays a log scale different colony forming unit count. PDA with chloramphenicol demonstrates the highest concordance with qPCR of ITS3 and whole genome sequencing. DRBC under counts yeast and mold. PDA without selection overcounts CFUs due to bacterial growth on PDA without selection.
