METHODOLOGICAL INFORMATION Most of the methods are standard and well described in doi: 10.3390/pharmaceutics14030602, doi: 10.1021/acs.biomac.8b00924,doi: 10.3390/pharmaceutics15041197. For in vivo studies, the fluorescence accumulation in mice tumor tissue of T22-GFP-H6 in different formats was measured at day 1 and at 10. Mice presenting tumors of approximately 120 mm3, randomized in three groups: control or buffer solution, T22-GFP-H6 IN material (soluble format) and T22-GFP-H6 Microparticles formed by Zn-mediated. The samples were subcutaneously injected to the corresponding mouse. Following, we measured the fluorescence accumulation in the remote tumor tissues . This would allow comparing the protein released pattern between the IN soluble protein and the MPs microgranules. For Size exclusion chromatography a quantity of 0.5 mg of OUT and protomer format of T22-GFP-H6 was injected into a Superdex 200 increase 10/300 GL (Cytiva) and performed in an ÄKTA pure system (GE Healthcare) after the equilibration with carbonate buffer with salt (166 mM NaCO3H, 333 mM NaCl, pH 8). The elution was performed with the same buffer at a flow rate of 0.5 mg/ml.
